Dinutuximab for the treatment of pediatric patients with high-risk neuroblastoma.
Neuroblastoma (NB) is the most common extra cranial solid tumor of childhood, with 60% of patients presenting with high risk (HR) NB by means of clinical, pathological and biological features. The 5-year survival rate for HR-NB remains below 40%, with the majority of patients suffering relapse from chemorefractory tumor. Immunotherapy is the main strategy against minimal residual disease and cl...

Purpose: In 2010, a Children's Oncology Group (COG) phase III randomized trial for patients with high-risk neuroblastoma (ANBL0032) demonstrated improved event-free survival (EFS) and overall survival (OS) following treatment with an immunotherapy regimen of dinutuximab, GM-CSF, IL2, and isotretinoin compared with treatment with isotretinoin alone. Dinutuximab, a chimeric anti-GD2 monoclonal an...

Purpose GD2 is highly expressed in neuroblastoma (NB) and specifically recognized by antibodies ch14.18. Immunotherapy with bolus infusion of ch14.18 in combination with cytokines prolonged survival in patients at risk for relapse but was associated with substantial toxicity, in particular severe pain, requiring i.v. morphine [EJM 2010]. In an attempt to reduce the pain side effect associated w...
